Abstract:
Most existing gene sequencing methods require a separated step of DNA template enrichment during sample preparation before sequencing, leading the library preparation process to be labor intensive and time consuming. Single molecule targeted sequencing (SMTS), enables direct sequencing of individual DNA molecules without any prior enrichment, overcoming the limits of most NGS approaches. SMTS utilizes wide-field total internal reflection fluorescence (TIRF) microscopy to simultaneously detect a large array of single molecule signals emitted from fluorescently labeled nucleotides. Exquisitely designed TIRF illumination only excites fluorescence within a shallow layer of evanescent wave above the flow cell surface and rejects unwanted background noise from the sample medium, yielding excellent signal-to-noise ratio for imaging single molecules. We have built a single molecule gene sequencer with two-color TIRF optics and developed image processing algorithms to accurately locate and identify DNA single molecules for bioinformatic analysis. SMTS with gene-specific flow cells expects to become a new platform extremely suitable for cancer gene mutation detection, infectious disease detection, inherited condition screening and noninvasive prenatal diagnosis.

Biography:
Dr. Zhiliang Zhou earned his BS in Instrumentation Science from University of Science & Technology of China (USTC) in 2006, and finished his joint PhD program in USTC & CAS (Chinese Academy of Sciences) in 2012. After that, Dr. Zhou worked in the Key Laboratory of Computational Optical Imaging of CAS, where he was leading the lab’s research on the field of light field imaging and developed a wide variety of computational imaging devices, including light field cameras, snapshot spectral-polarimetric imager, super-high dynamic range telescope, etc. After joining Direct Genomics in 2015, Dr. Zhou is now dedicating himself to R&D of single molecule fluorescence imaging systems for the 3rd generation gene sequencers. His team has successfully developed the first single molecule sequencer in Asia, receiving extensive attention worldwide.
